چکیده

Purpose: Cobalt-chloride (CoCl2) induces a strong degeneration of the porcine retina through hypoxic mechanisms. Neurons of the inner retina layers are particularly affected. This model is used to study possible protective effects of hypothermia treatment. Methods: Organotypic cultures of porcine retinas were cultured and treated with 300 μM CoCl2 for 48 h. Hypothermia (30°C) was applied for the same duration. Results: At day 8, RGC layer thickness decreased. In accordance, a loss of retinal ganglion cells (RGCs) was noted in the CoCl2 group (p = 0.0004), while RGCs were rescued through hypothermia. The amount of apoptotic RGCs was higher in the CoCl2 group (p = 0.0002), but also present in the CoCl2+hypothermia group (p = 0.002). Conclusions: A protection of RGCs could be achieved through hypothermia application in a hypoxia porcine retina organ culture. This alternative model is suitable for drug and treatment screening and will therefore reduce the number of animal studies. VII-29

Lutein Protects RGC-5 Cells Against Hypoxia and Oxidative Stress

Retinal ischemia and oxidative stress lead to neuronal death in many ocular pathologies. Recently, we found that lutein, an oxy-carotenoid, protected the inner retina from ischemia/reperfusion injury. However, it is uncertain whether lutein directly protects retinal ganglion cells (RGCs). Here, an in vitro model of hypoxia and oxidative stress was used to further investigate the neuroprotective...

Hypothermia protects human neurons

BACKGROUND AND AIMS Hypothermia provides neuroprotection after cardiac arrest, hypoxic-ischemic encephalopathy, and in animal models of ischemic stroke. However, as drug development for stroke has been beset by translational failure, we sought additional evidence that hypothermia protects human neurons against ischemic injury. METHODS Human embryonic stem cells were cultured and differentiate...

P-123: The protective Effects of The Vitamin E on Sperm DNA Damage Induced by Mobile Phone Radiation

Background: Cobalt (Co) is an essential trace element for mammals required for the synthesis of vitamin B12. It is not a cumulative toxin but chronic exposure induces negative effects on the organism. It was proven that cobalt passes via placenta appearing in the fetal blood and amniotic fluid and it is shown to possess an embryotoxic effect. Also, chromium (Cr) is recognized as a trace element...

Baclofen Protects Primary Rat Retinal Ganglion Cells from Chemical Hypoxia-Induced Apoptosis Through the Akt and PERK Pathways

Retinal ganglion cells (RGCs) consume large quantities of energy to convert light information into a neuronal signal, which makes them highly susceptible to hypoxic injury. This study aimed to investigate the potential protection by baclofen, a GABAB receptor agonist of RGCs against hypoxia-induced apoptosis. Cobalt chloride (CoCl2) was applied to mimic hypoxia. Primary rat RGCs were subjected ...

Lutein Attenuates Both Apoptosis and Autophagy upon Cobalt (II) Chloride-Induced Hypoxia in Rat Műller Cells

Retinal ischemia/reperfusion injury is a common feature of various retinal diseases such as glaucoma and diabetic retinopathy. Lutein, a potent anti-oxidant, is used to improve visual function in patients with age-related macular degeneration (AMD). Lutein attenuates apoptosis, oxidative stress and inflammation in animal models of acute retinal ischemia/hypoxia. Here, we further show that lutei...
